Update it, so Professor Fate is a woman. More to the point, instead of Fate and a prince, it's now Fate and a princess.

You'd need someone who can play two roles.

Tatiana Maslany (if you've seen "Orphan Black," you know what I'm talking about)

This would mean she'd get twice as messy.

Never seen "The Great Race" so I have absolutely no idea who they would play, but I would greatly appreciate it if Anne Hathaway and Sabrina Carpenter were in it getting completely pied, slimed, soaked, slopped and messy

I agree with the Professor Fate update. Make it a woman. There are a number of actresses have the acting chops who could pull it off Anne Hathaway, Meryl Streep, Cate Blanchett, Margot Robbie.
